Transcriptomic and metabolomic analysis reveals the effect of hydrogen sulfide on delaying fruit ripening and color transition of white fleshed strawberry (Fragaria ananassa)

摘要: Strawberry (Fragaria ananassa) is a globally popular and economically important horticultural crop. However, ripe strawberry fruit is highly perishable and vulnerable to microbial contamination. Our previous studies have shown that hydrogen sulfide (HAS) regulates the ripening of postharvest white fleshed strawberry, but the regulatory mechanism remains unknown. In this study, sodium hydrosulfide (NaHS) treated samples showed no significant color alteration during ripening, while hypotaurine (HT) treated samples exhibited significant color alterations at the initial ripening (S2) and full ripe (S3) stages. Meanwhile, NaHS treated fruit showed higher firmness and total anthocyanin content, yet lower levels of total soluble solid and sucrose at S3 stage. Endogenous HAS content increased in the NaHS treated group but decreased in the HT-treated group, with similar trends shown in the activities of L-cysteine desulfhydrase and D-cysteine desulfhydrase and the expression levels of FaLCD1 and FaDCD2. Untargeted metabolomic analysis identified 832 secondary metabolites and classified them into 55 classes. Metabolite enrichment analysis revealed that 'Phenylpropanoid biosynthesis' pathway played a role in SW fruit ripening process. Transcriptomic analysis revealed the role of key DAMs in fruit maturation, showing pathways like 'Phenylpropanoid biosynthesis' were important throughout, and 'Starch and sucrose metabolism' at S3 stage. Co-expression network analysis revealed 62 transcription factors potentially regulating phenylpropanoid synthesis genes (Fa4CL3, FaF3H3, FaPAL2) under NaHS and HT treatments, with Fa4CL3 positively correlated with ERF13, FaF3H3 with NAC031, and FaPAL2 with MYB75. In addition, yeast one-hybrid and dual-luciferase assays suggested that FaERF13 was directly bound to the promoters of Fa4CL3 and FaF3H3. Overall, this study sheds light on the precise regulatory mechanism of H2S regulating strawberry ripening process, which provides potential insights for the research on the cultivation and post-harvest storage of strawberries.
