摘要: Monitoring nitric oxide (NO) release at single cell level is fundamental to understand the diverse physiological/pathophysiological functions of this remarkable signal molecule. Here, a new versatile and selective analytical method using capillary electrophoresis with laser-induced fluorescence detection was originally developed for monitoring NO release in individual cells after choosing 8-(3,4-diaminophenyl)-2,6-bis(2-carboxyethyl)-4,4-difluoro-1,3,5,7-tetramethyl-4-bora-3a,4a-diaza-s-indacene (DAMBO-P-H). The high resolution quality of this method permit to quantify NO release in single cells quickly, reproducibly and selectively, yielding a sensitive approach to in situ single-cell analysis with a detection limit reaching attomole level. The feasibility of the method was fully validated by successfully applied to measurement of NO release in four single cell study models for the understanding of the pathobiological and therapeutic basis of NO for cardiovascular diseases, and for the study of the virus infection of high pathogenicity avian influenza virus (H5N1), HBV, HCV and human influenza virus A, respectively.
