Observations on the formation and development of the primary germinal tissue of cultured Chinese sturgeon, Acipenser sinensis

摘要: Formation and development of primary germinal tissue of cultured Acipenser sinensis were studied by light microscope. The primordial germ cell was firstly seen between the nephridium area and the yolk sac of the eleutheroembryos, at about 1.7cm TL (third day after hatching). By the 11th day, the genital fold formed, which was composed of several cell layers. With the primordial gonad tissue growing, the blood vessels appeared when fish were 2 months old (average TL of 7.1 cm). The adipose fatty tissue appeared at the age of 7 months (average TL 59 cm). Histological indications for sex differentiation in the gonads occurred at the age of 9 months (0.6 similar to 1.1kg body weight (BW), while the gonads entered the maturity stage 1 during which mitosis of the spermatogonia and oogonia was first observed. With the primary spermatocytes and oocytes appearing, the testes and ovaries reached stage 2 (in males at an age of about 22 to26 months, (1.55 to5.6 kg BW.), while the ovaries reached maturity stage 2 when fish were 30 to36 months old(3.1 to8.2 kg BW). After the age of 3 years, the gonads, in succession, reached a stage that could be sexed in most cases by gross examination, and at the the age of 5 years (18 to35.5kg BW), all of the gonads can be easily sexed by gross examination while the oocyte diameter showed a range in diameter between 60 similar to 240 mu m (physiologically stage 2). Besides, from the results on the developmental sequence it can be inferred that in the Yangtze River, all of the fingerlings migrating downstream may arrive in the estuary with too little gonad development that would already allow gross anatomical sex differentiation and most of the specimens will be so immature that conventional histological studies would also not permit sex differentiation.
