The unnoticed risk: Chronic nitrite exposure disturbs the intestinal barrier, new insights from juvenile starry flounder (Platichthys stellatus)

摘要: The intestinal mucosal barrier is a vital defense system that integrates physical, chemical, immunological, and microbial defenses to maintain host health. However, the response of this barrier to chronic nitrite exposure remains poorly understood. Thus, the study delved into the implications of sustained nitrite exposure on the functionality and integrity of the intestinal mucosal barrier in Platichthys stellatus. After a 30-day exposure period, NO2-N (0.4 mg/L and 0.8 mg/L) breaks the intestinal physical barrier, as evidenced by altered histopathology of the intestinal epithelial and reduced expression of tight junction (TJ) genes (claudin-like, occludin, and ZO-1). The chemical barrier was compromised, indicated by a thinning mucus layer and a loss of goblet cells. Additionally, inflammatory responses were triggered, with elevated pro-inflammatory cytokines (TNF-alpha, IL-8, and IL-6) and reduced anti-inflammatory IL-10 levels, disruption to the intestinal immunological barrier. Furthermore, chronic nitrite exposure also disrupted microbial homeostasis, increasing Aliivibrio abundance and leading to higher alpha-diversity and network complexity in the microbiota. This microbial shift caused metabolic disturbances, impairing the expected functions of the intestinal microbiota. These findings highlight the severe impact of chronic nitrite exposure on the intestinal mucosal barrier of P. stellatus. This study is the first to provide evidence of such disruption in fish, offering crucial insights for managing nitrite levels in the intensive aquaculture of starry flounder.
