摘要: In this research, we developed a mixed agglutination reaction (MAR) assay based on core-shell Fe3O4@SiO2 magnetic nanobeads (MNBs) to detect IgG anti-sperm antibodies (AsAb) in human semen and analyzed the feasibility of this novel approach for evaluating male fertility. We prepared human IgG modified Fe3O4@SiO2 magnetic nanobeads (hIgG-MNBs) and applied these hIgG-MNBs for analyzing 5 semen samples, collected from the Guangxi Zhuang Autonomous Region of south China. For comparison, traditional IgG/A MAR tests, which based on latex microsphere carrier, were also conducted in parallel. Results obtained from the MNB MAR test were similar with those of traditional MAR tests; however, the percentages of positive sperms in MNB MAR tests were lower. During magnetic separation, Sperm Class Analyzer system was used to capture the traces of moving sperm-NMBs composites, which indicated that the bonding between sperms and MNBs was strong and stable to support the motion of sperm-MNBs composite under magnetic field. The hIgG-MNBs conjugates synthesized in this study showed an excellent biocompatibility, and the conjugates easily separated the sperms that contained anti-sperm antibodies; thus, it reduced the chances of false-positive results and contributed to the standardization of new method. By modifying the surface of nanoparticles with different probe molecules, they can be used for the separation of other biological entities, such as sperms and cells.
