OPTIMIZATION OF IN VITRO MICROSPORE CULURE TECHNIQUES IN BARLEY AND ITS APPLICATION TO BREEDING

摘要: The factors influencing survival of isolated microspores and production of embryo-like structure (ELS) were studied using field-grown plants. The results showed that 1) more survived microspores could be obtained from several-sowing method; 2)A numberof multi cellular structure (MCS) was produced by in vitro culture of isolated microspores derived from barley spikes pretreated at 3°C for 20 days and the production of MCS and ELS were significantly promoted by adding barley floret in to induction medium, especially adding floret derived from tetraploid plants of barley; 3) The regeneration of green-plants could be improved by early transfer of ELS. Stable-lines could be developed through platform of DH breeding within two years and stable variety could be developed within 4 years.
