摘要: In order to obtain an optimal transformation efficiency of Bacillus subtilis electroporation experimental program,to optimize the conditions for electroporation of Bacillus subtilis cells,a shuttle vector for Escherichia coli and Bacillus subtilis,pHY300PLK,were used.Factors including growth medium,electroporation medium,recovery Medium,plasmid concentration and pulse strength were investigated.Single factor experiments were employed to optimize the growth medium,electroporation medium,recovery Medium,plasmid concentration and pulse strength.Results showed that the optimal values for concentration of the growth medium electroporation medium,recovery Medium,plasmid concentration and pulse strength are LB+0.5 M sorbitol,0.5 M sorbitol+0.5 M mannitol+10% Glycero,LB+0.5 M sorbitol+0.38 M mannitol,20 kV/ cm and 100 ng/μl ofPlasmid DNA.
