Spatiotemporal relationship of wheat stripe rust occurrence revealing the key inoculum origin area and migration route of Puccinia striiformis f. sp. tritici in the Yunnan-Guizhou epidemiological region

摘要: Aerial urediniospores of Puccinia striiformis f. sp. tritici (Pst), the causative agent of wheat stripe rust, can be transported across nearly all wheat-growing regions of China throughout the year. Recent studies have shown that the Yunnan-Guizhou epidemiological region (YGER) plays a significant role in providing inoculum that contributes to disease outbreaks in the major wheat-producing areas of eastern China. However, the temporal dynamics of Pst inoculum on wheat and the migration route of the rust within the YGER remain unclear. To address this, we conducted an intensive investigation into the occurrence of stripe rust on wheat and collected 388 isolates from 12 wheat production regions of the YGER from October 2021 to December 2022. All isolates were phenotyped using 18 Yr single-gene differential lines, along with 8 additional differential lines, and genotyped using a genotyping by targeted sequencing with Pst134E as a reference genome to determine the migration route of the rust in this region. The results demonstrated that the early-season wheat cultivation zones in the north-central YGER serve as key inoculum sources due to the early onset of the disease, higher diversity, rich ancestral composition, and close relationship of Pst with other regions. Temporal analyses of disease occurrence and subpopulation relationships revealed that inoculum from the north-central YGER is transmitted to the southwestern, southern, and eastern. Therefore, inoculum from the north-central YGER is primarily responsible for the spring wheat stripe rust epidemics within the YGER. These findings provide valuable insights into the epidemiology and integrated management of wheat stripe rust in the YGER.
