Krüppel-homolog 1 is key in development and parthenogenetic reproduction in pea aphids

摘要: Kr & uuml;ppel-homolog 1 (Kr-h1) is a key juvenile hormone (JH)-induced transcription factor and plays an important role in insect development and reproduction. However, the molecular function of Kr-h1 in aphids is still unknown. In this study, we used the parthenogenetic pea aphid, Acyrthosiphon pisum, to investigate the presence and role of ApKr-h1 in postembryonic development and reproduction. The tissue distribution and developmental profiles demonstrated that ApKr-h1 was highly expressed in 4th-instar nymph and adults and embryo. Knockdown of ApKr-h1 in 4th-instar nymph resulted in a precocious molting and 79% of aphids were arrested in their molting process. The rest aphids that were able to successfully develop into adults failed to sclerotize normally and died rapidly without producing offspring. Hence, RNA-seq and functional assays revealed that ApKr-h1 can directly regulate the expression of a cuticle protein ApCP53 by binding to its promoter, potentially explaining the failure of molting. Moreover, injection of adults with dsApKr-h1 significantly decreased the fecundity of A. pisum. Our findings not only enrich our knowledge on the function of the Kr-h1 and JH in the development with molting process and reproduction of A. pisum, but may also provide potential targets for RNAi-based management of aphids employing JH-related genes.
