The Effects of Different Hormones and Concentration of Major Element Stock Solution on the Proliferation of in Vitro Grapevine

摘要: Tissue culture is hardly affected by season and the seedlings are neat and consistent, which meets the needs of short-term large-scale production. In this study, the shoot tips and stem segments of six in vitro grapevine plantlets were used as original materials. A total of 12 types of media formulations were set up to explore the effects of different hormones and concentration of major element stock solution in MS medium on in vitro 'Tamina', 'Merlot' and 'Beda' plantlets. The results showed that the height of in vitro plantlets cultured in half major element MS medium was shorter than that in the whole nutrient medium. The change of 6-benzylaminopurine (6-BA) concentration had a certain inhibition effect on the rooting of the plantlets, but the regularity was not obvious. The change of kinetin (KT) concentration had no obvious effect on proliferation, plant height and rooting. After two rounds of subculture, two types of media (No. 6 and No. 12) showing better proliferation were chosen and the shoot tips and segments of 'Fenghou', 'Kyoho', 'SO4', '101-14', 'Cabernet Sauvignon' and 'Riesling' were used for further verification. The results showed that the proliferation of in vitro plantlets in No. 6 medium is better than that in No. 12 medium. Among the six varieties, the shoot tip and stem segment of 'Riesling' and the shoot of two rootstocks, '101-14' and 'SO4', had lower proliferation, less than 2, and the other varieties was more than 2. The proliferation of stem section per plantlet for 'Fenghou', 'SO4', and 'Cabernet sauvignon' was the highest, up to 6. In conclusion, the stem segment of in vitro grapevine was suitable material for proliferation and the efficiency medium 1/2MS + 0.4 mg/L 6-BA + 0.5 mg/L KT + 0.2 mg/L IAA + 30.0 g/L sucrose + 6.0 g/L agar (pH 5.8).
