Molecular characteristics of three complete genome sequences of apple rubbery wood virus 1 isolates from apple trees in China

摘要: Apple rubbery wood virus 1 (ARWV-1, genus Rubodvirus, family Phenuiviridae) is a causal agent of rubbery wood disease, and is also associated with flat limb disease in apple. ARWV-1 has been detected in apple trees in China, however, its complete genome sequences remain unreported. Here, we report the complete nucleotide sequences of three isolates of ARWV-1 (FJ, XH, and XF) collected from apple trees in Xingcheng, Liaoning Province, China. Their genomes contain three RNA segments: L, M, and S. Sequence alignment showed that the genomic sequences of FJ, XH, and XF have 94.3-99.6%, 93.6-99.8%, and 84.5-99.8% nucleotide identity, respectively, with the five previously reported ARWV-1 isolates. Phylogenetic analyses based on the nucleotide sequences of the L, M, and S segments indicated that all ARWV-1 full genome isolates could be divided into three clades. In phylogenetic trees for each genome segment, isolates FJ, XH, and XF formed a clade with isolate BR-Mishima from Brazil. This is the first report of complete genome sequences of ARWV-1 from apple trees in China. Our results will provide important reference information for the prevention and control of this virus.
