Cloning and characterization of one novel PR1 gene from Coptis chinensis responding to southern blight

摘要: Southern blight, caused by Sclerotium rolfsii, is a major threat to Coptis chinensis production, but the plant's defense mechanisms and resistance genes are poorly understood. In this study, transcriptomic analysis of healthy and infected C. chinensis identified the key resistance gene Coptis chinensis pathogenesis-related protein 1 (CcPR1). The full sequence of CcPR1 was cloned and analyzed bioinformatically. qRT-PCR showed that CcPR1 expression was significantly upregulated in pathogen-infected plants, highlighting its role in disease resistance. Additionally, anti-fungal assays of the recombinant CcPR1 protein from Escherichia coli confirmed its activity. Subcellular localization placed the protein in the extracellular matrix. CcPR1 overexpression in Nicotiana benthamiana leaves significantly inhibited S. rolfsii and Botrytis cinerea growth, suggesting broad-spectrum resistance to fungal pathogens. This study advances the understanding of CcPR1 and its role in C. chinensis defense against southern blight.
