Development of molecular markers for total soluble solids in tomato fruits

摘要: Total soluble solids (TSS) is the primary determinant of flavor and taste during the development and ripening of tomato fruits. Unfortunately, genetic loci and molecular markers for identifying tomato cultivars with high TSS contents are currently scarce. In this research, the metabolome profiles of red ripe fruits from nine F1 tomato cultivars were measured to identify those with superior flavor and higher TSS content, and two tomato cultivars WTS21-3 and WIP21-9 with higher taste values and TSS contents were further analyzed. Through bulk segregant analysis (BSA), we performed whole-genome sequencing of the F2 segregation population of the two tomato cultivars WTS21-3 and WIP21-9. Association analysis disclosed that the variation in TSS concentration was correlated with a C -> T base mutation in SNP1 (SL4.0ch01_85381466). Therefore, molecular markers were developed for the SNP1 site. In the F2 population of tomato cultivars WTS21-3, the selection efficiencies of the TSSSNP1 and SNP1KASP markers in high TSS lines were 86.7 % and 83.33 %, while in low TSS lines, the marker selection efficiencies were 66.7 % and 44.43 %, respectively. This study provides new genetic materials and molecular markers for the selection and improvement of tomato cultivars with high TSS.
