Changes in the Metabolome of Flue-Cured Tobacco by Dioctyl Phthalate and Erucamide: Mechanism of Continuous Monocropping

摘要: The allelopathic effect is one of the most important mechanisms of continuous cropping obstacles. In previous works, dioctyl phthalate (DP) and erucamide (ER) were demonstrated to be correlated with cropping years. In the present study, the effects of DP and ER on the plant transcript profiling and metabolome were studied to reveal the potential mechanism. Seed germination experiment was primarily conducted with 20 mg/L, 25 mg/L, and 30 mg/L DP or 50 mg/L, 100 mg/L, and 150 mg/L ER to screen concentration for pot experiment. The pot experiment was performed with tobacco seedlings aged 45 d treated with 30 mg/L DP or 150mg/L ER. The growth indexes, transcriptome, and metabolome of flue-cured tobacco were analyzed to evaluate the effect and mechanism of DP and ER on tobacco growth and development. Both DP and ER significantly inhibited seed germination and suppressed the growth and development of flue-cured tobacco. The transcript profile revealed metabolic pathways and biosynthesis of secondary metabolites were closely correlated with the changes in flue-cured tobacco. Further metabolome results revealed that sulfur metabolism was involved in the effect of DP on tobacco, while the effect of ER was mainly related to alpha-linolenic acid metabolism, plant hormone transduction, and lysine biosynthesis. DP and ER significantly affected the metabolism and secondary metabolites biosynthesis of tobacco, which suppressed the growth and development of flue-cured tobacco, and therefore induced continuous cropping obstacles. This study could provide a theoretical basis for the prevention and control of continuous monocropping obstacles.
