High polymorphism and moderate differentiation of chub mackerel, Scomber japonicus (Perciformes: Scombridae), along the coast of China revealed by fifteen novel microsatellite markers

摘要: Genetic diversity and population structure of Scomber japonicus along the coast of China (from Yellow Sea-YS to East China Sea-ECS and South China Sea-SCS) were analysed in 319 individuals from 11 sites using 15 novel microsatellite loci. Relatively high levels of genetic variation within population (Na = 10.20-13.20, Ho = 0.507-0.686, He = 0.678-0.805, and PIC = 0.669-0.776) and moderate level of genetic differentiation among populations (F_(ST) = 0.1055) were detected. UPGMA, PCA and Bayesian analyses suggested that S. japonicus from YS to SCS can be divided into three major groups: one including all populations from YS extending to the eastern and western coast of Hainan Island in SCS, the second consisting of one population off the southern coast of Hainan Island in central SCS, and the third comprising one population in middle ECS. Furthermore, we inferred that there was at least one spawning stock of S. japonicus in YS, and the larvae/juveniles of S. japonicus tended to migrate from YS southward to SCS. Wilcoxon signed-rank test and mode-shift indicator of allele frequency distribution support that S. japonicus had not undergone recent genetic bottlenecks. This study revealed the population genetic diversity and structure of S. japonicus, and will be useful to the sustainable utilization, effective management and conservation of S. japonicus resource in the future.
