Effects of culture media, carbon and nitrogen sources and environmental factors on mycelial growth and sporulation of Pestalotiopsis microspora strains, the agent of bayberry twig blight in southern China

摘要: The effects of media types, carbon (C), nitrogen (N) sources, temperature, and pH on Pestaliopsis microspora, a pathogen of bayberry blight, were examined to determine optimal conditions for mycelial growth and light on sporulation. Three independent isolates showed a similar trend of growth and sporulation with different C and N sources. However, the growth rates varied significantly among seven agar media. Fungal growth rates were highest on potato dextrose agar (PDA), potato sucrose agar (PSA), and bayberry fruit extract agar (BFA); moderate on oatmeal agar (OMA) and Czapek-Dox agar (CDA), and low on bayberry leaf extract agar (BLA) and bayberry branch extract agar (BBA). Among the different sources of carbon, sucrose, dextrose, maltose, lactose, mannose, levulose, sorbose and starch exhibited good effects on growth rate and denseness of the three isolates; glycerol and ribose were optimal for growth rate but moderate for denseness; and xylose was poor for the isolate YS26, but moderate for YS44 and RA1-2. Urea was good for YS44 and RA1-2, but moderate for YS26. Potassium nitrate, ammonium nitrate and all of the amino including cysteine, glycine, tyrosine, histidine, alanine, arginine, glutamate and proline were good sources of nitrogen for isolate YS26, but alanine, glutamate and proline and tyrosine were moderate for YS44 and RA1-2, respectively. Growth occurred between 10-30 degrees C and the optimal temperature was between 25-28 degrees C for YS26 and YS44 and 25 degrees C for RA1-2. Growth of the three isolates was good and had no significant difference between pH 5 to 10. The quantity of conidia from YS26 on PDA at 5 days growth under light was significantly greater (p < 0.001) than under darkness. In contrast, sporulation of the other two isolates, YS44 and RA1-2, was unaffected by light. These results will be helpful for understanding the epidemiology of the bayberry twig blight disease and for developing a reasonable disease management strategy in reforestation areas.
