Gibberellin application and RNA-seq-based transcriptomtc analysis of leaf and buds in grape

摘要: A tetracyclic diterpenoid hormone, Gibberellins are a class of plant hormones critical in controlling several plant growth and development processes, including fruit ripening, flower production, stem elongation, and seed germination. However, the effect of gibberellin on grape leaves and bud development is not yet reported. In this study, we found that GA3 could release bud dormancy, and increase the GA3, and IAA content, but decrease ABA content. GA3 also increased the chlorophyll content, and SOD, POD, CAT, and APX activities to promote bud germination and leaf development. RNA-Seq was used to analyze grape bud and leaf information to obtain many differentially expressed genes (DEG) and study the general changes in response to gibberellin. Results showed that based on readings per million base reads per million base map reads (RPKM) values, we obtained 2025 differential genes common to leaf and bud combinations. Analysis by gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) showed that after gibberellin spraying, the accumulation and metabolism of carbohydrates, defense functions, and coloration of grape leaves and buds were affected. In addition, gibberellin might promote the photosynthesis of grape leaves and buds and the open response center of the light system. Gibberellin interacts with salicylic acid, as demonstrated by the down-regulation of the salicylic acid pathway and the activation of the DELLA protein (GAI1) in plant hormone signaling. This study established a strong basis for further investigations into the molecular process behind the formation of grape buds and leaves.
