Enzymatic and histopathological changes during resistance of cowpea to Fusarium oxysporum f. sp tracheiphilum

摘要: We evaluated a resistant cultivar ('Fengchang No. 2') and a susceptible cultivar ('Zhijiang-28-2') of cowpea (Vigna sesquipedalis) for changes in the activities of defense enzymes after challenging seedlings with Fusarium oxysporum f. sp. tracheiphilum. Phenylalanine ammonia-lyase, beta-1,3-glucanase, chitinase, peroxidase, polyphenol oxidase, and catalase activities in hypocotyl tissues were analyzed. We also studied the infection process in the primary xylem, using light microscopy. There appeared to be deposits in the vessels and thickening of the vessel walls. To further elucidate the resistance mechanisms, the susceptible cultivar, 'Zhijiang-28-2', was treated with salicylic acid, ferulic acid, and oxalic acid, then the susceptibility to the pathogen and the beta-1,3-glucanase and chitinase activities were determined. Treatment with salicylic acid at 0.5 mmol/L was most effective in reducing infection levels. Our work suggests an important involvement of a set of enzymes in resistance of cowpea to F. oxysporwn f. sp. tracheiphilum.
