Anthocitrus: evaluation of anthocyanin accumulating "Mexican" lime fruits produced by overexpressing the Ruby transcription factor gene from Citrus sinensis "Moro"

摘要: Anthocyanin accumulation naturally occurs in the 'blood orange' (Citrus sinensis (L.) Osbeck) and is controlled by a retrotransposon that is inserted in the promoter of the Ruby transcription factor upon cold stress. Several enzymes work sequentially for anthocyanin biosynthesis following the activation of Ruby. We analyzed the juice vesicles of non-transgenic and the transgenic 'Mexican' lime that overexpressed the Ruby transcription factor to understand the regulatory mechanism of anthocyanin biosynthesis in transgenic fruits. Cyanidin and petunidin anthocyanins were detected using liquid chromatography in the juice extracted from Ruby transgenic 'Mexican' limes. Additionally, we identified a positive correlation between the overexpression of anthocyanin and the total content of flavonoids and phenolic compounds. In contrast, overexpression of Ruby had no significant effect on the ascorbic acid content or DPPH scavenging capacity of transgenic juice. Several volatile compounds were also detected at a high concentration in the transgenic juice. Many of the anthocyanin biosynthesis-related genes, including PAL, CHS, CHI, F3H, F3 & PRIME;H, DFR, and ANS, were upregulated along with the upregulation of MYB113, WD40, bHLH, and R2R3-MYB complex-related genes in transgenic juice vesicles. In contrast, the expression of the negative regulators of anthocyanin biosynthesis were downregulated including SQUAMOSA PROMOTER BINDING PROTEIN-9 (SPL9) and S-LIKE RIBONUCLEASE (RNS1). Our results provide pertinent information on genetically modified anthocyanin-rich citrus fruits with enhanced phenolic and flavonoid compounds resulting from the induction of phenylpropanoid and anthocyanin pathways. Key messageAnthocyanin overexpressing transgenic "Mexican' Lime fruits have enhanced phenolic and flavonoid compounds that may provide beneficial to human health and wellbeing.
