Optimization of Catechin Nanoliposomes and Evaluation of Their Antioxidant Activity and Cytotoxicity

摘要: Catechin, which is a major component in green tea polyphenols, has been demonstrated to be a strong antioxidant. In this work, the effectiveness of catechin was enhanced by using liposomes. The response surface methodology was successfully used to optimize the biochemical and biotechnological processes. Optimal preparation conditions were as follows: phosphatidylcholine-to-cholesterol molar ratio: 3.13; catechin concentration: 3.83 mg/mL; and rotation speed: 30 rpm. Particle size and encapsulation efficiency were 221 +/- 4 nm and 76%+/- 1%, respectively. The modified surface structure of the optimized catechin nanoliposomes was observed by scanning electron microscopy, which showed that the prepared nanoliposomes were approximately 200 nm large, and had regular spherical shape. The antioxidant activity of the catechin nanoliposomes was evaluated. Results showed that the antioxidant activities of catechin nanoliposomes for the scavenging of 2,2'-azinobis-3-ethylbenzthiazoline-6-sulfonic acid, hydroxyl, and 1,1-diphenyl-2-picrylhydrazyl radicals in vitro was obviously improved. These results indicated that the liposomes protected the catechins from the influence of the external environment. Cytotoxicity over Caco-2 cells was assessed and catechin nanoliposomes were cytotoxic (a significant difference was assessed, compared with the blank control) only at high concentrations (0.025 mg/mL). In addition, higher cytotoxicity was found at 36 h than at 24 h at the same concentrations.
