Cloning and expression analysis of key genes for lignin synthesis in Cucurbita pepo L.

摘要: Cucurbita is one of the oldest domesticated crops and is widely cultivated worldwide. Hull-less Cucurbita is a natural mutant which was discovered in normal Cucurbita. Hull-less seed has the potential values in snack food, functional food and further processing to extract the vegetable oil. Hull-less Cucurbita, an excellent germplasm resource, has a substantial market demand. Lignin is a major component of plant cell walls and is regarded as an important factor affecting seed coat formation. PAL (phenylalanine ammonia lyase), 4CL (4-coumaric acid coenzyme A ligase), CCR (Cinnamoyl-CoA reductase), CAD (Cinnamyl alcohol dehydrogenase) and C4H (Cinnamate-4-hydroxylase) play key roles in lignin biosynthesis in plants, and the full-length sequences of these genes were cloned from Cucurbita pepo L. seed coats by PCR, exhibiting open reading frames of 2136, 1632, 2373, 1083 and 1518 bp, respectively. Bioinformatics analysis showed that the proteins encoded by the corresponding genes were highly conserved in Cucurbitaceae. Transcription analysis results indicated that these detected genes had higher relative expression in hulled C. pepo than in hull-less C. pepo during testa development. With seed coat development, the gene expression first increased and reached a peak from 10 to 20 days, except for CpCAD, and then decreased gradually. These results revealed that these genes were differentially expressed in both genotypes and were closely related to lignin synthesis in testa, eventually affecting seed coat formation. Thus, this study provides a theoretical foundation for better utilizing and developing excellent cultivars of hull-less C. pepo.
