Transformation of the rolB gene via the pollen-tube pathway to obtain transgenic pomegranate plants

摘要: To enhance the efficiency of gene transformation and accelerate genetic improvement of pomegranate strains, the Agrobacterium rolB gene was transferred into 'Tunisia' soft-seeded pomegranates via the pollen-tube pathway method and successfully expressed. Transferred exogenous DNA showed only minor mechanical damage to the embryo and no obvious influence on fruit. Furthermore, expression of rolB gene was reproduced in transgenic plants after four cycles of culture, indicating the stability of this method. The rolB-transferred plants had a higher number of roots and greater root lengths compared to control, indicating that they were more likely to take root. Therefore, transgenic pomegranates were successfully obtained, demonstrating the potential application of the pollen-tube pathway method in pomegranate breeding programs to obtain new cultivars with favorable agronomic traits.
