Characterization of the Galectin-3 gene and its association with Streptococcus iniae resistance traits in Acanthopagrus latus (Houttuyn, 1782)

摘要: Galectin-3 (Gal-3) is the only chimera-type member of the galectin gene family and belongs to the pattern recognition receptor family. It has been proven that it plays an important role in innate immunity in teleost. This study aimed to characterize Gal-3 within the whole genome of the yellowfin seabream, Acanthopagrus latus, and investigate its role in the resistance to Streptococcus iniae infection. The genomic sequence of the A. latus Gal-3 gene (AlGal-3) spanned 10,910 bp, encompassing 8 exons and 7 introns, with an mRNA length of 1236 bp, and featured a carbohydrate recognition domain (CRD). Molecular docking analysis of AlGal-3 suggested that it could bind to N-acetylglucosamine at a conserved carbohydrate-binding motif within the CRD. Quantitative PCR assays in healthy tissues revealed the highest expression levels observed in the stomach and gills, significantly higher than in other tissue, and followed by the spleen, head kidney, and intestines. Upon infection with S. iniae, differential upregulation in AlGal-3 expression was observed in the brain, liver, spleen, and head kidneys. Furthermore, we identified three single nucleotide polymorphisms within the AlGal-3 gene. We selected the first 80 individuals that died and the last 80 individuals that did not die of 2000 infected yellowfin bream individuals, to serve as the susceptible group or resistant group. Notably, the single nucleotide polymorphisms at position 10362G/A in the seventh intron was significantly associated with resistance to S. iniae infection. Our findings provide a foundation for the selection of disease resistance traits in yellowfin seabream and offer new insights into the role of Gal-3 in the immune recognition mechanisms of teleosts.
