摘要: Blind-side hypermelanosis in flounder is a serious issue in aquaculture. Gynogenesis, a haploid breeding technique that significantly enhances genetic homogeneity and inherits specific traits, contributes to the elucidation of molecular mechanisms and improvements in breeding. In this study, we used recombinant double haploid (rDH) populations of normal and hypermelanotic Japanese flounder ( Paralichthys olivaceus) ) generated by gynogenesis, skin transcriptome libraries, and segregation pools (Po_N and Po_H) for BSA-seq. Based on a combination of RNA-seq and BSA-seq, eight loci were identified, including five InDels and three SNPs. Genotyping of 204 individuals from diverse rDH and wild populations revealed the presence of three loci within ca4a and mreg, , with ca4a exhibiting significant enrichment in nitrogen metabolic pathways. qRT-PCR results revealed significant differences in the relative expression of ca4a and mreg between pigmented blind-side skin of hypermelanotic Japanese flounder, blind-side skin of normal Japanese flounder, and non-pigmented blind-side skin of hypermelanotic Japanese flounder, indicating that their candidate genes are involved in the formation of blind-side hypermelanosis in Japanese flounder. The identification of candidate differentially expressed genes (DEGs) and loci will provide important genetic resources for analyzing the molecular mechanisms of blindside hypermelanosis and for breeding improvement in Japanese flounder.
