摘要: The recombinant plasmid containing ovary aromatase gene from BAC library of Nile tilapia, Oreochromis niloticus, was extracted and purified according to the instruction of QIAGEN Plasmid Mini Handbook with a little of modification. The probe of aromatase gene for the in situ hybridization was prepared from the plasmid DNA and labelled with biotin by degenerate oligonucleotide primed PCR (DOP-PCR). The labelled probe was then applied in the fluorescence in situ hybridization (FISH) to the mitotic metaphase chromosomes of Nile tilapia in order to localize the aromatase gene in the Nile tilapia. The result showed that the ovary aromatase gene of Nile tilapia was located on one pair of small chromosome other than the sex chromosome. So the aromatase gene is not primary sex determining gene in Nile tilapia, but its function to sex indicates that some autosomal loci can affect sex determination and differentiation in fish.
