A universal PCR primer for detection of members of the potyviridae

摘要: The Potyviridae is the largest of the plant virus families currently recognized. This family consists of 6 genera based on their transmission by fungi (Bymovirus), whiteflies (Ipomovirus), aphids (Macluravirus, Potyvirus) or mites (Rymovirus, Tritimovirus). All members have filamentous particles 650 approx 900 nm or 500 approx 600 and 200 approx 300 nm in length and 11 approx 13 nm in width, made up of a single structural coat protein surrounding a linear, single-stranded positive sense monopartite or bipartite RNA genome. All the viruses induce characteristic pinwheel or scrollshaped inclusion bodies in the cytoplasm of the infected cells. Because the genome organization of different members is similar, and some regions of genomes among different members are high conserved, it is possible to develop universal primers for use in RT-PCR to detect these viruses from infected plant materials. In this paper, a universal primer was designed from a highly conserved motif in the NIb domain and RT-PCR used to amplifty the 3'-terminal sequences of different members of th family. In addition, 3'-terminal sequences of oat necrotic mottle virus (ONMV) and celery mosaic virus (CeMV) were determined for the first time and a potyvirus infecting maize in China was identified.
