基于RNA-seq筛选影响猪肉品质的关键候选基因

摘要: 本研究旨在探究影响从江香猪和大白猪肉质差异的关键候选基因.实验以90日龄的从江香猪和大白猪为对象,利用转录组(RNA-seq)技术分析2个猪种背最长肌中差异基因表达谱,对差异表达基因(DEGs)进行GO和KEGG富集分析.qRT-PCR进行验证和筛选出肉质调控的关键功能基因.结果表明:生物信息学分析筛选出2个猪种DEGs 318个,其中上调基因有184个,下调基因有134个;GO分析发现,富集最显著的前30个GO条目,主要参与生物过程和细胞组分功能;KEGG分析发现,富集最显著的4条通路均与肉质相关,包括一磷酸腺苷活化蛋白激酶信号通路(AMPK Signaling Pathway)、转录因子信号通路(FoxO Signaling Pathway)、丙酮酸代谢(Pyruvate Metabolism)和脂肪酸代谢途径(Fatty Acid Metabolim).通过筛选与验证,得到影响2个猪种肉品质的关键候选基因,其中同属硒蛋白家族的硒蛋白W(SelW)和硒结合蛋白1(SBP1)在2个品种猪中具有不同的表达模式,SelW在大白猪中高表达,而SBP1在从江香猪中高表达.本研究基于RNA-seq技术揭示了大白猪和香猪背最长肌中基因表达谱的差异,筛选肉质形成相关的信号通路和关键功能基因,为猪肉品质的分子调控机制奠定了基础.
