Effects of arginine supplementation in high-carbohydrate diets on the growth, hematological parameters, and hepatic and skeletal muscle glucose metabolism of juvenile mirror carp ( Cyprinus carpio) ) based on PI3K/Akt signaling pathway

摘要: High-carbohydrate diets can lead to decreased growth performance and disrupted glucose metabolism in fish. As arginine has the function of improving glucose absorption, the purpose of this study was to investigate the effects of arginine on the growth performance and glucose metabolism of mirror carp ( Cyprinus carpio) ) in a high- carbohydrate diet. Mirror carp (6.84 +/- 0.02 g) were assigned into five diet groups: high-carbohydrate diets (45 % starch level) supplemented with 0 (A0), 0.6 % (A0.6), 1.2 % (A1.2), 1.8 % (A1.8), and 2.4 % (A2.4) arginine. The results of an 8-week trial period revealed that adding more than 1.2 % arginine to the high- carbohydrate diets significantly increased the final body weight, weight gain rate, and protein efficiency ratio, while decreasing the feed conversion ratio. A high-carbohydrate diet containing more than 1.8 % arginine significantly reduced serum triglyceride concentrations, while the A2.4 group had the lowest serum cholesterol. The activity of glucokinase (GK), 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 2 (PFKFB2) and pyruvate kinase (PK) in the liver was significantly increased in the A2.4 group, while the activity of phosphofructokinase (PFK) in the liver was notably decreased. The activity of GK, PK and PFKFB2 in the muscle was significantly elevated in the experimental groups supplemented with more than 1.2 % arginine. Supplementing appropriate levels of arginine significantly raised the levels of phosphatidylinositol-3-hydroxykinase (PI3K), serine/threonine protein kinase (Akt), and p-Akt in the liver and muscle. Furthermore, adding appropriate levels of arginine to the high-carbohydrate diets increased the expression of genes associated with glucose transport (protein kinase B substrate 160, glucose transporter 4), glycolysis (PFKFB2, GK, and glycogen synthase), and glycogen synthesis (glycogen synthase), while inhibiting the expression of genes related to gluconeogenesis (glucose-6-phosphatase and phosphoenolpyruvate carboxykinase). The results indicated that supplementing appropriate levels of arginine (1.2 % and 1.8 %) to the high-carbohydrate diets could enhance the growth performance of mirror carp. This enhancement was probably achieved by regulating the expression of genes linked to glucose metabolism in the PI3K/Akt signaling pathway.
