An efficient Agrobacterium tumefaciens-mediated transformation system of Taraxacum kok-saghyz Rodin (Russian dandelion)

摘要: Taraxacum kok-saghyz Rodin (TK) is an excellent alternative to Hevea brasiliensis because of its high yield of latex, wide growth range, and short life cycle, and it is also an ideal plant for natural rubber research through genetic transformation. This study aimed to report an efficient Agrobacterium tumefaciens-mediated transformation system of TK using leaves as explants. A TK genotype, named 12-16, that is suitable for tissue culture was obtained from a wild germplasm repository in Xinjiang. The multiplication rate of TK genotype 12-16 reached 3.72 adventitious shoots per explant through cultivation on the shoot induction medium of Murashige-Skoog (MS) nutrient medium supplemented with 1.5 mg L-1 6-benzylaminopurine and 0.1 mg L-1 naphthaleneacetic acid. Moreover, the average length of adventitious shoots was 3.07 +/- 0.78 cm after 60 d in culture. Based on the transformation system, two genes of rubber elongation factor 138 (REF138) and rubber elongation factor 258 (REF258), driven under an enhanced 35S promoter, were transformed into the genotype 12-16 through Agrobacterium tumefaciens GV3101 mediated genetic transformation. The transformation process from leaf explants to hygromycin-resistant seedlings lasted approximately 3 to 4 mo, and the transformation efficiency reached 23.12 to 24.05%. This study established a highly efficient transformation system based on the TK genotype 12-16 and provided an important technical support for rubber biosynthesis research.
