摘要: Algal-associated bacteria are fundamental to the ecological performance of marine brown macroalgae such as Saccharina japonica. As an economically important species, the success of its field aquaculture is highly dependent on the epiphytic microbiota associated with health. However, the structure and assembly of the epiphytic bacterial community (EBC) on S. japonica have been scarcely investigated, notably the temporal changes during the field cultivation period. Here, the diversity, structure, and assembly pattern of the EBC on S. japonica meristems were investigated monthly from January to June during the large-scale cultivation period. The phylogenetic richness of the EBC decreased from January to April and then increased from April to June. The variation in EBC structure was highly dependent on sampling time. The classes gamma-Proteobacteria, Actinobacteria, alpha-Proteobacteria, and Bacteroidia predominated in the EBC, with 17 core members affiliated with these groups. Both environmental factors and the growth of S. japonica contributed to the variation in the EBC, and 11 identified indicator members were significantly associated with them. Deterministic processes played significant roles in community assembly notably during the mushroom stage, i.e., from February to April, and then gradually weakened, resulting in a stochasticity-dominated community by June. This study provides information on the temporal assemblage and underlying ecological mechanisms of the meristematic EBC associated with S. japonica, which is critical to establishing baseline data on a "healthy" microbiome for future aquaculture of this species.
