Furunculosis in Farm-Raised Brachymystax lenok in China

摘要: Furunculosis which frequently occurs in farm-raised Brachymystax lenok has seriously affected the healthy development of cold-water fish aquaculture in China. This study focused on the pathogenic characteristics of furunculosis in order to provide a theoretical basis for diagnosis, prevention, and control of this disease in B. lenok. One strain of the causal organism, Aeromonas salmonicida subsp. salmonicida, named LN1301 was isolated from the liver of diseased juvenile B. lenok showing typical symptoms of septicemia and characteristic histological changes in liver, spleen, kidney, and intestine of fish suffering from furunculosis. An infection experiment using two means of inoculation was carried out with healthy juvenile B. lenok. Results showed that LN1301 was highly pathogenic to juvenile fish and was characterized by the occurrence of hemorrhages on the base of fins and in several other organs. LD50 value of LN1301 to fish was determined to be 2x10(5) CFU/mL and 4.5x10(6) CFU/mL respectively when the fish were inoculated by intraperitoneal injection and immersion of experimentally wounded fish respectively. Bacteria that were re-isolated from the artificially infected fish showed the same characteristics as the LN1301 strain, which was proved to be a pathogenic form of A. salmonicida which caused furunculosis. Compared to similar sequences in the GenBank database, 16S rRNA sequence of LN1301 was 99% identical with A. salmonicida subsp. salmonicida. Antimicrobial susceptibility tests indicated that LN1301 strain was highly susceptible to 9 kinds of drugs but resistant to gentamicin, amoxicillin, ampicillin, carbenicillin, and rifampicin.
