Integrative Metabolomic and Transcriptome Analysis Reveal the Differential Mechanisms of Spot Color in the Lips of Dendrobium chrysotoxum

摘要: Dendrobium chrysotoxum is a beautiful and valuable ornamental orchid. The lip of flowers usually contains an orange spot, but purple-spot germplasm is also found in wild resources, and the differential mechanisms of spot colors are still unclear. In this study, orange spot (OS) and non-orange spot (NOS) from cultivated plants, purple spot (PS) and non-purple spot (NPS) from wild plants were used as research materials, and compared through tissue section, metabolomics and transcriptome analysis. The examination of transversal section showed purple pigments accumulated in the glandular hairs of upper epidermis of PS, yellow pigments accumulated in all cell layers of OS, NOS and NPS. A total of fifteen carotenoids were identified, and phytoene,alpha-carotene, beta-carotene and gamma-carotene accumulated in large quantities in OS, and the total carotenoid content in OS reached 2981.78 mu g/g, which was 2.63, 4.15 and 3.12 times than that of PS, NOS and NPS, respectively. Fifteen anthocyanins were identified and PS contained the highest content with three unique anthocyanins. In PS, upregulated expression of CHS, CHI, F3H, F3 ' H, F3 ' 5 ' H, ANS and 3AT genes promoted anthocyanin accumulation compared with OS. While OS had high levels of phytoene, alpha-carotene, gamma-carotene and beta-carotene owing to high expression of upstream genes (PSY, PDS, Z-ISO, ZDS and LCYB) and down-regulated downstream genes (LUT1, BCH, LUT5, ZEP and VDE). The downregulation of carotenoid structural genes in NOS is also the main reason for the color difference between OS and NOS. Eighteen and seventeen transcription factors related to anthocyanin and carotenoids were screened, respectively. This results revealed the differential mechanisms of spot color in the lips of D. chrysotoxum, which would be useful to provide a reference for similar research in other orchids.
