Bile acids help maintain immune homeostasis and enhance antioxidant capacity upon Vibrio infection in leopard coral grouper

摘要: The leopard coral grouper, Plectropomus leopardus, is an economically important marine fish. However, frequent occurrences of diseases have severely limited the sustainable development of its farming industry. Bile acids (BAs) play a crucial role in regulating glycolipid metabolism, and immune responses; however, the molecular mechanisms underlying its role in regulating the fish immunity and disease resistance remain illusive, which has largely hindered their application in aquaculture. In this study, leopard coral groupers were fed with BAs and infected with its major bacterial pathogen Vibrio harveyi. A combined analysis of transcriptomics and metabolomics revealed that, the supplements of BAs elevated the taurochenodeoxycholic acid (TCDCA) and glycocholic acid (GCA) levels in liver, which resulted in an upregulation of choline metabolites and downregulation of linoleic acid (LA) and arachidonic acid (AA). The upregulation of choline metabolites enhances the expression of anti-inflammatory factors, such as interleukin-10 receptor beta (il-10rb), thereby mitigating the host's intense inflammatory response. Concurrently, the downregulation of LA and AA, reduces the production of reactive oxygen species (ROS). Furthermore, the elevated levels of 10-formyltetrahydrofolate and the increased expression of glutathione synthetase (gss) gene may promote glutathione synthesis, indicating an improved antioxidant capacity. These results suggest that BAs help maintain host immune homeostasis and alleviate excessive oxidative stress to cope with the Vibrio infection. These findings provide a theoretical foundation for elucidating the molecular mechanisms underlying the role of BAs in regulating the immune response, which could contribute to the control and management of diseases in the aquaculture of the leopard coral grouper.
