摘要: With the rise of metabolites/molecules production using complete living cells or their components (bacteria, enzymes, chloroplasts,.) which is otherwise named as bioprocess, it has become increasingly important to control the different production steps and to analyse the different biological materials .Among its complete products and technical portfolio, Horiba Scientific can provide instruments especially dedicated to therapeutic cell manufacturing processes. On one hand, spectroscopic-based systems such as fluorescence and Raman can be implied at the stage of upstream processes for cell culture analysis. Fluorescence spectroscopy was shown to be a viable and rapid method of identifying and evaluating the quality of cell culture media.Using Raman spectroscopy combined with microscopy and multivariate analysis, it has been shown that different species and growth phases of single bacteria cells can be quickly distinguished.On the other hand, Surface Plasmon Resonance imaging (SPRi) and Particle Size Analysis (PSA) techniques can be of high interest for downstream processes in the characterization of the components produced by the living organisms. SPRi with its multiplex feature and label-Free approach is a very useful technique to rank and screen molecules such as antibodies produced by hybridoma technique or phage display technology. For such applications, SPRi will allow an easy and fast selection of hybridoma cell lines according to affinity or kinetic rate parameters of the produced monoclonal antibodies to their specific antigen. High throughput screening is the best way to accelerate the validation and selection of molecules while reducing both time and costs .As an example, PSA technique can be used to control the final formulated product which corresponds to the purified product mixed with several inert ingredients as excipients.
