Identification and functional analysis of a novel single nucleotide polymorphism (SNP) in the porcine Toll like receptor (TLR) 5 gene

摘要: Toll like receptor (TLR) 5, recognizing bacterial flagellin, plays an important role in immune systems. Single nucleotide polymorphisms (SNPs) in TLR5 gene may alter host immune response to flagellin and have an effect on susceptibility to infectious disease, as suggested by studies in humans and other animals. Here, a novel nonsynonymous SNP, G2239A leading to the transformation of Val by Met at the 747th amino acid, was first identified in TIR domain of porcine TLR5 gene. To examine whether SNP G2239A is functionally relevant, eukaryotic expression vector with site-directed mutagenesis at the site was constructed and its ability to activate the expression of TLR5-dependent reporter constructs was tested. Variant notably diminished the activation of reporter gene (p < 0.05) when compared with wild type TLR5. SNP G2239A has an effect on the receptor function.
