Highly Efficient Gene Transfer with HIV-1-based Vectors in Cells in vitro

摘要: We have optimized a protocol for the bulky preparation of HIV-1-based defective lentiviral vector (DLV) and the concentration of vector through one-step ultracentrifugation. DLV demonstrated high transduction efficiency in human T-cell lines. Long-term culture of transduced CEM-ss cells detected by genome DNA PCR and total RNA RT-PCR showed a stable expression of reporter green fluorescent protein (GFP) gene without apparent alternation in cellular morphology and growth kinetics. 11 cell lines were tested for the susceptibility to DLV infection. Highly efficient gene transfer was revealed in CEM-ss, MT-2, Sup-T1 and HTB-14 cells. DLV is one of the promising gene transfer vehicles in the development of human gene therapy protocols.
