Assessing Genetic Diversity and Genetic Structure of Three Yellowfin Seabream Populations in Chinese Waters Using the AFLP Technique

摘要: The yellowfin seabream Acanthopagrus latus H. is one of the primary and important aquaculture species in southeastern China. In this study, three wild populations (n=118) of the yellowfin seabream from Xiamen (XM), Shenzhen (SZ), Beihai (BH) were analyzed using the AFLP technique. Four primer sets revealed a total of 244 putative loci. Of which, 119 were polymorphic (48.7%). The proportions of polymorphic loci of the three populations varied from 29.5% (population XM) to 36.5% (population BH) with an average of 32.3%, while the Nei's gene diversities (Hj) ranged from 0.20 (S.E.0.02; population SZ) to 0.22 (S.E.0.01; population BH) with an average of 0.21 (S.E.0.01). So, there existed distinct variations among populations. Furthermore, both the genetic distance analysis and cluster analysis clustered population XM and population SZ in a group, while population BH as a single cluster. The Wright's Fst analysis also suggested there existed significant genetic differentiation among populations (F st=0.0275; p=0.0003), especially between population BH and populations of XM and SZ (p < 0.01). It therefore would be proper to consider the populations of XM and SZ to be one geographic stock, while the population BH belongs to another geographic stock.
