摘要: Transgenic techniques have become the new approach for developing scab-resistant wheat germplasm. We have successfully developed a stable and rapid transformation system for wheat using a particle-bombardment method. Immature embryos or scutella, 13-14 days after pollination, were used as a recipient and the bar gene was the selectable gene. The gold particle density and distance from the stop screen to the target tissue were the important factors influencing the transformation efficiency. The most satisfactory results were obtained with 42 mg of particles/bombardment. If inmiature embryos were given an osmotic treatment 4 hours before and 16 hours after bombardment, the transformation efficiency would increase. Presently, we are planning to transfer antifungal genes, such as chitinase, 6-1,3-glucanase, and the defensin gene, to common wheat in hopes of obtaining new germplasm with scab resistance.
