Cloning of disease resistance-relevant Nramp and MHC IIB genes and their application potential in molecular breeding for disease resistance in Japanese flounder (Paralichthys olivaceus)

摘要: Nramp and MHC IIB genes were isolated from Japanese flounder (Paralichthys olivaceus) using PCR and RACE-PCR. RT-PCR indicated that Nramp transcripts were highly abundant in spleen, headkidney, abundant in intestine, liver and gill, and less abundant in heart. Significant association between MHC IIB polymorphism and diseases resistance / susceptibility was found in Japanese flounder. 13 different MHC IIB alleles were identified among 411 clones from 84 individuals. Most alleles such as allele a, b, c, d, e, f j, k, i, m were commonly found in both resistance stock and the susceptibility stock. However, the allele g and h were observed only in resistance stock, and allele 1 was only found in susceptibility stock.
