摘要: Apple chlorotic leafspot virus (ACLSV) and apple stem grooving virus (ASGV) are two important viruses of apple and other fruit crops. They infect many commercial apple cultivars with infection rate of up to 80-100% and cause yield loss of up to 40% in P. R. China. Protein A sandwich enzyme-linked immunosorbent assay (PAS-ELISA) has been used as a routine method for the detection of ACLSV and ASGV and for screening of virus-free materials generated from virus elimination programs. However. this method involves more steps and thus time consuming. The standard and modified version of the double antibody sandwich ELISA (DAS-ELISA) were compared with PAS-ELISA for their effectiveness in detecting ACLSV and ASGV from tissue culture of commercially important cultivars. Results indicated that the standard DAS-ELISA was successful to detect ASGV but failed to detect ACLSV. The modified DAS-ELISA showed similar efficacy to PAS-ELISA in detecting both viruses.
